Short-period circadian rhythm in constant light. From Supplementary Figure 3 of Gould et al. 2006: "The gi mutants gi-611 and gi-596 were isolated from an EMS-mutagenised population carrying the CAB:LUC+ transgene in the WS background, in a genetic screen for plants with altered temporal regulation of CAB:LUC+ expression [see Kevei et al., The Plant Journal, 2006]. Genetic mapping (>90 chromosomes) showed that each mutation was tightly linked to GI; sequencing revealed G-A transition mutations in the GI coding region in both lines, leading to predicted amino acid changes L281?F in gi-611 and S191?F in gi-596. Description Using mRNA null alleles, we investigated the role of FT and TSF in flowering. The single mutants showed no significant impact, but the double mutant displayed photoperiod-insensitivity. Furthermore, this double mutant fully suppressed early flowering caused by CO overexpression. Biochemical assays revealed similar functions for FT and TSF, supported by their interaction with identical bZIP transcription factors in yeast. Additionally, early flowering was induced in the double mutant by expressing FT or TSF from phloem companion cell-specific promoters, indicating no necessity for gene expression in the meristem.

Initially, we identified putative binding targets through computational analysis of SOC1 regulatory sequences and known flowering-time regulatory genes. Subsequent ChIP-seq assays confirmed the binding of SOC1 to its own locus as well as to various flowering-time regulatory and floral homeotic genes. In planta analyses revealed the repression of SOC1 by floral homeotic MADS domain proteins, dependent on the presence of SOC1 itself. Mechanistic investigations further elucidated the molecular basis of this regulatory interplay. Overall, our study provides insights into the complex regulatory mechanisms governing flowering time and floral development in Arabidopsis thaliana.
